Can I export data from an .est file (Microsoft S&T 2009) with any GPS tool?

Yes Larry, you can. You can open your Microsoft's Streets & Trips .EST files to edit them, or to save for use with any other popular GPS brand (Garmin, Magellan, Nokia, iPhone 3G, etc. ) or software.
I use http://garmin.gps-data-team.com/poi_manager.php for that.
I can easily make a dedicated POI, EST to CSV, EST to GPX, EST to OV2 or EST to KML converter if needed.

I have MS Streets & Trips but can't seem to find the .EST files. Where are they located?

When you save a file in Microsoft Streets & Trips, the saved file will have .est extension. The new Streets & Trips 2010, by default, saves in your Documents folder, but of course you may opt for a different location to save your files on your computer.
